Preparation of Thiolated Chitosan (TC)/Astragalus Membranaceus Oil (AM) Microcapsules by Sonochemical Method

Abstract: Traditional Chinese herb has attracted extensive attention due to its good efficacy, low toxicity and low side effects. However, some active ingredient extracts are relatively sensitive to external influence and lose their effectiveness. Microcapsules, as a new type of drug delivery carrier, can encapsulate drugs and improve the bioavailability. After several decades of development, microcapsules can be prepared through many synthetic methods. But there are some problems to be solved, such as the complicated preparation process, the introduction of harmful substances, etc. Compared with conventional microencapsulation methods, sonochemistry is simple, green and efficient. In this paper, Chitosan as raw material and 2-iminothiane hydrochloride as sulf hydrylation reagent were used to prepare thiolated chitosan (TC). Using TC as the shell and astragalus membranaceus oil (AM) as the core, we therefore obtained TC/AM core-shell microcapsules by sonochemistry. The effects of thiol content and pH value on the synthetic reaction were investigated. The optimal degree of thiolation and pH value were determined through energy spectrometer (EDS), laser confocal scanning microscope (CLSM), dynamic light scatterer (DLS), etc. The results showed that the optimal mass ratio of chitosan and thiolation reagent was 5∶1, and the pH value was 6.0. The particle size of TC/AM microcapsules prepared under the optimal condition were characterized by DLS and consequently was 962.4 nm.
