Controlled Release Properties of a Time controlled/pH dependent Colonic Drug Delivery System for Berberine Hydrochloride |

Abstract The present work proposed and successfully prepared a time-controlled/pH dependent colonic drug delivery system for berberine hydrochloride (BH), i.e. BH/MCM-41/CS-SA, by a combinative method of oscillation absorption and dip-coating, using MCM-41 mesoporous silica molecular sieve as the main body, BH drug molecules as the object, natural polymers — sodium alginate (SA) and chitosan (CS) as the coating materials. The physical and chemical properties of BH/MCM-41/CS-SA were characterized by XRD, SEM, BET and FTIR, and its drug loading and release properties in vitro were evaluated by spectrophotometry. The results showed that the drug loading rate of BH/MCM-41/CS-SA was 23.5%, and the mesoporous structure of MCM-41 was not destroyed after loading. In vitro results indicated the conspicuous time-delay/pH dependent drug release characteristic of BH/MCM-41/CS-SA compared with BH/MCM-41, which displays both favorable colon specific drug delivery performance and long-term controlled release efficacy.
