Abstract Bridged organic-inorganic hybrid mesoporous silica, also referred to as periodic mesoporous organosilicas (PMOs), is a kind of mesoporous material that has covalently incorporated organic moieties into the silica framework using controlled synthesis techniques. Unlike standard mesoporous silica materials, which are purely inorganic, PMOs use both inorganic and organic properties and advantages having mechanical strength, thermal stability, highly ordered pore architecture, tunable pore size distribution, biocompatibility and low cytotoxicity;these properties indicate that PMOs are prime candidates for delivery of drugs and nanomedicine applications. This review categorizes PMOs on the basis of brid-ging groups, functionalization techniques, describes selected contemporary advances in the synthesis and functionalization of PMOs, and outlines design considerations and current status of stimuli-responsive drug delivery using PMOs that respond to pH, redox, hypoxic, and photothermal (i.e.heat) stimuli as possible tumor therapies. The intention of this review is to serve as a reference guide for rational design of functional PMOs and next generation drug delivery platforms.
